It shouldn't be any problem what I now of. The limitations are within sql express itself. I think the system requirements are written as it is from a "political" point of view rather than technical. Meaning that if you have a large enterprise site you cannot be expected to get support from us to tweak episerver cms to run on a sql express database that has as you say memory restrictions, which could be a customer argument for not saying they cannot use sql express.
In otherwords we don't recommend it and don't support it but go ahead at your own risk...

Since Episerver 5.0 r2 supports SQL Server 2008 express when developing / testing - can it also run on SQL Server 2008 express in production environment?
There are some limitation to the database such as max 4 Gb database, 1 Cpu and some memory restrictions, but does EpiServer require some features that are not included in the express version?
